Drug dosing was limited by the occurrence ofadverse effects. Apparent tolerance to adverse effects was observedin two subjects when the dose of physostigmine was escalated slowlyover at least 2 weeks. Steady-state cholinesterase inhibition byphysostigmine appears to produce sustained cognitive improvement insome subjects with Alzheimer disease without substantially alteringits therapeutic index.
