L-dopa carbidopa treatment of senile dementia: a control study

A study has been carried out on the effect of L-Dopa in Alzheimer's disease. A total of 21 patients, who had a DSM- III-R of Alzheimer's disease, were randomly assigned to two groups of 13 and 8 patients. The first group was treated with a combination of L-Dopa-Carbidopa (500 and 50 mgs per day), and the second with placebo, for nine months. Two raters evaluated each patient, prior to the beginning of treatment and each month thereafter. Of all the items only tremor, labile affect and speech were found to improve in a statistically significant way. Rigidity also improved while the level of medication was low, but at higher dosages it reappeared. This effect may be related to the adaptation of the dopaminergic receptors.
